Cardarine is a common name for GW501516, a synthetic compound developed in the 1990s through research collaborations involving GlaxoSmithKline. It belongs to a class of molecules known as peroxisome proliferator-activated receptor delta agonists. Early studies explored its effects on lipid metabolism and energy expenditure in animal models. The compound was never approved as a human medicine, and clinical development was discontinued. In the years since, it has appeared in fitness and bodybuilding communities as a performance-enhancing substance. Regulatory agencies classify it as an unapproved drug.
PPARδ is a nuclear receptor that regulates gene expression related to fatty acid oxidation, glucose homeostasis, and mitochondrial function. GW501516 binds to this receptor with high affinity and activates downstream signaling in skeletal muscle and other tissues. Animal studies reported increased endurance and altered fuel preference, but human data remain limited and inconsistent. The precise relationship between receptor activation and observed physiological changes is still an area of active investigation. Researchers have also examined whether the compound affects inflammation or cell proliferation. No approved therapeutic indication exists for cardarine.
In laboratory settings, cardarine is studied as a tool compound for probing PPARδ biology. Published experiments often use cell cultures, rodent models, or isolated tissues. Some investigations focus on metabolic effects, while others assess potential risks such as carcinogenicity observed in long-term animal studies. Because human trials are sparse, most knowledge comes from preclinical work and adverse event reports. Scientific literature frequently notes the gap between animal findings and human outcomes. The compound is not a dietary supplement and is not intended for human consumption.
Laboratory studies have examined GW501516 in cell cultures and rodents for conditions such as dyslipidemia, insulin resistance, and obesity. Some trials in humans were initiated, but development was discontinued after preclinical findings raised concerns about cancer in certain models. Those findings do not prove that the compound causes cancer in people, but they contributed to regulatory caution. Later reviews often describe the evidence as preliminary and insufficient for assessing long-term safety.
In the fitness and bodybuilding literature, cardarine is frequently discussed as an endurance agent or fat-loss compound, although such claims are not supported by robust clinical evidence. Online descriptions often mix animal data, user anecdotes, and marketing language. Researchers who study PPARδ agonists distinguish between receptor activation in controlled experiments and unsupervised use of unverified products. The latter introduces unknown purity, dose, and interactions, making reported experiences difficult to interpret scientifically.
GW501516 acts as an agonist at peroxisome proliferator-activated receptor delta, a nuclear receptor involved in transcription of genes related to lipid handling and energy use. Activation of PPARδ can shift skeletal muscle toward greater fatty acid oxidation in animal models, which is one reason it drew interest for metabolic disease and exercise research. The exact downstream effects depend on tissue, species, dose, and duration. Human data are sparse, so many proposed benefits remain hypotheses rather than established clinical outcomes.

Regulatory bodies treat GW501516 as a prohibited substance in competitive sport. The World Anti-Doping Agency added it to the prohibited list, and it falls under classes covering metabolic modulators and hormone-related agents. It is not approved by drug regulators for human use, and it is not a lawful dietary supplement. Products sold under the cardarine name may contain unlisted ingredients or different compounds. Because no approved product exists, quality and identity are not guaranteed by pharmaceutical manufacturing standards.
Cardarine is a common name for the investigational chemical GW501516, also written GW-1516. It was developed as a peroxisome proliferator-activated receptor delta agonist for metabolic conditions such as dyslipidemia. Early research focused on lipid handling and energy use in skeletal muscle and other tissues. The compound was never approved as a medicine. In public discussion, it is often grouped with performance-enhancing substances, although its receptor target differs from that of anabolic steroids or selective androgen receptor modulators. Regulatory and health authorities have issued warnings about its use.
GW501516 acts on PPARδ, a nuclear receptor that helps regulate fatty acid oxidation and energy homeostasis. In animal studies, activation of this receptor was associated with increased endurance and changes in lipid metabolism. Human trials examined effects on blood lipids and other metabolic markers, but the compound did not advance to approval. Rodent studies later reported tumors in multiple tissues at doses used in those experiments. Whether those findings translate to human risk remains uncertain, and the clinical relevance of the animal data is still debated.
Safety concerns emerged from long-term animal studies. In rodents given the compound for extended periods, researchers found an increased incidence of certain cancers, including liver and bladder tumors. These findings contributed to the discontinuation of clinical development. Whether similar risks apply to short-term or low-level exposure in humans is not established, and controlled human safety data are limited. The relevance of high-dose rodent carcinogenicity findings to human use remains a subject of debate.
Human trials of GW501516 were small and short in duration. They examined lipid levels, glucose handling, and other metabolic markers, but the programs were halted after the animal cancer findings. No approved therapeutic product exists, and published human data are insufficient for establishing long-term safety. Reports of use for athletic performance come mainly from non-clinical settings and cannot be verified through controlled trials. Independent testing of products sold as cardarine has found inconsistent purity and labeling.
== History and etymology == Valine was first isolated from casein in 1901 by Hermann Emil Fischer. The name valine comes from its structural similarity to valeric acid, which in turn is named after the plant valerian due to the presence of the acid in the roots of the plant.
Glu-270 and Arg-127 play an important role in catalysis shown in Figure 2. Arg-127 acts to stabilize the carbonyl of the substrate that is bound to amino group of phenylalanine. Simultaneously, the water molecule coordinated to zinc is deprotonated by Glu-270 and interacts with the carbonyl stabilized by Arg-127. This creates an intermediate, shown in Figure 2, where the negatively charged oxygen is coordinated to zinc, and through unfavorable electrostatic interactions between Glu-270 and the ionized product facilitates the release of the product at the end of catalysis. In recent computational studies, the mechanism of catalysis is similar but the difference in mechanism is that deprotonated water molecule binds to the carbon of the carbonyl, whereas Figure 2 shows the hydroxyl group stays coordinated to zinc. Then proteolysis occurs and the water molecule is then introduced back into the active site to coordinate to zinc. Several studies have been conducted exploring the details of the bond between carboxypeptidase A and substrate and how this affects the rate of hydrolysis. In 1934, it was first discovered through kinetic experiments that, in order for substrate to bind, the peptide that is to be hydrolyzed must be adjacent to a terminal free hydroxyl group. Also, the rate of hydrolysis can be enhanced if the C-terminal residue is branched aliphatic or aromatic. However, if the substrate is a dipeptide with a free amino group, it undergoes hydrolysis slowly; this, however, can be avoided if the amino group is blocked by N-acylation.
The first 7-Eleven store in Denmark was opened at Østerbro in Copenhagen on September 14, 1993. There are 185 stores, mostly in Copenhagen, Aarhus, Aalborg, and Odense, including two stores at Copenhagen Central Station. In Denmark, 7-Eleven had an agreement with Shell, with a nationwide network of Shell/7-Eleven service stations. This ended in 2024 when the national franchise operator Reitan Danmark did not renew the agreement and instead used their own brand of service station Uno-X. 7-Eleven also have an agreement with the Danish railway company DSB to have 7-Eleven stores at most S-train stations and other train stations. In 2022, 7-Eleven in Denmark suffered a widespread ransomware attack that caused all stores to temporarily close. 7-Eleven did not comply with the attacker's demands. No customer data was compromised in the attack. The three biggest 7-Eleven stores in terms of revenue in the world, are all located in Denmark. The top two stores are both located at Copenhagen Central Station and the third are located at Copenhagen Airport beyond security.
Phosphate analysis reveals high concentrations of cremations in the barrows, in the form of satellite and secondary burials in the round barrows. This suggests that the primary burials may have been of chiefs, or socially powerful/respected people, and that some people may have paid to be buried close to the person they respected or followed.

=== Murder of Charmaine West === Rose is believed to have murdered Charmaine shortly before Fred's prison release date of 24 June 1971. She is known to have taken Charmaine, Anne Marie and Heather to visit Fred on 15 June, and it is believed that Charmaine was killed on or very shortly after this date. As well as forensic dentistry confirming that Charmaine had died while Fred was still incarcerated, further testimony from Tracey Giles' mother, Shirley, corroborated the fact that Charmaine had died before Fred's release. In her later testimony at Rose's trial, Shirley stated that her two daughters had been playmates of Charmaine and Anne Marie when her family lived in the upper flat of 25 Midland Road in 1971. Shirley further stated that after her family had vacated the flat in April 1971, she had brought Tracey to visit Charmaine on a day in June, only for Tracey to be told by Rose: "She's gone to live with her mother, and bloody good riddance!" Shirley Giles was adamant that Fred was still in prison on this occasion. As with the Giles family, Rose explained Charmaine's disappearance to others who enquired about her whereabouts by claiming that Fred's first wife, Catherine "Rena" West, had taken her eldest daughter to live with her in Bristol. She contradictorily informed staff at Charmaine's primary school that the child had moved with her mother to London. When Fred was released from prison on 24 June, he allayed Anne Marie's concerns for her sister's whereabouts by claiming Rena had collected Charmaine and returned to her native Scotland.
Conservation strategies include the maintenance of: 1) refuge plants and reservoir hosts to preserve the EcM fungal community after harvesting; 2) mature trees to provide seedlings with a diverse array of EcM fungi; and 3) old-growth stands that have diverse macro- and microhabitats and support varied EcM fungal communities. Preservation of natural forest floor constituents and retention of woody debris and substrates may also be important. In one study concerning Douglas-fir seedlings, removal of forest floor debris and soil compaction decreased EcM fungal diversity and abundance by 60%. Removal of pinegrass similarly reduced the diversity and richness of EcM fungi. Some strategies, such as prescribed burns, have different effects on different types of EcM communities, ranging from negative to neutral or positive. Large ex situ culture collections of fungi, including ectomycorrhizal fungi, are maintained throughout the world as insurance against genetic loss. However, these collections are incomplete.
On June 16, 2001, Sierra canceled the Dreamcast port of Half-Life, citing "changing market conditions". A late build of the Dreamcast version eventually leaked online, featuring complete versions of Half-Life and Blue Shift. Blue Shift and the High Definition pack were initially absent from the launch of Valve's content delivery system Steam in September 2003, despite the presence of both Half-Life and Opposing Force on the system. The game was released on Steam on August 29, 2005, along with the High Definition Pack. Blue Shift was also published as part of Sierra's Half-Life: Generation compilation in 2002, and as part of Valve and Electronic Arts' Half-Life 1: Anthology on September 26, 2005.
=== Septic arthritis === PCT at a cutoff value of .5 ng/mL was effective at ruling in septic arthritis in an analysis of over 8000 patients across 10 prospective studies. PCT had a sensitivity of 54% and specificity of 95%. The study also concluded that PCT outperforms C-reactive protein in differentiating septic arthritis from non-septic arthritis.
=== Drugs === Research and development in this field has been driven largely by the desire to find better cancer treatments. Tumors cannot grow larger than 2mm without angiogenesis. By stopping the growth of blood vessels, scientists hope to cut the means by which tumors can nourish themselves and thus metastasize. In addition to their use as anti-cancer drugs, angiogenesis inhibitors are being investigated for their use as anti-obesity agents, as blood vessels in adipose tissue never fully mature, and are thus destroyed by angiogenesis inhibitors. Angiogenesis inhibitors are also used as treatment for the wet form of macular degeneration. By blocking VEGF, inhibitors can cause regression of the abnormal blood vessels in the retina and improve vision when injected directly into the vitreous humor of the eye.

=== Ethidium bromide === Ethidium bromide intercalates and stains DNA, providing a fluorescent red-orange stain. Although it will not stain healthy cells, it can be used to identify cells that are in the final stages of apoptosis – such cells have much more permeable membranes. Consequently, ethidium bromide is often used as a marker for apoptosis in cells populations and to locate bands of DNA in gel electrophoresis. The stain may also be used in conjunction with acridine orange (AO) in viable cell counting. This EB/AO combined stain causes live cells to fluoresce green whilst apoptotic cells retain the distinctive red-orange fluorescence.
== Prognosis == As of March 2020, only 50 cases of CMRD have been documented in the medical literature. This small number speaks to the rarity of the disease as well as the lack of thorough research and documentation. As a result, the full course of the disease, life expectancy, and mortality are also poorly documented. Clinical manifestation of CMRD symptoms begin during infancy and early childhood but may go undetected due to the non-specific symptoms associated with the disease. Many of these symptoms can be attributed to malnutrition and nonspecific postnatal diarrhea, confounding early diagnosis. Careful regulation of diet and nutrition are required for management of CMRD since the disease results from the poor absorption of nutrients from food.
== Worldwide incidence by country == The total number of centenarians in the world is uncertain. The Population Division of the United Nations estimated that there were 23,000 in 1950, 110,000 in 1990, 150,000 in 1995, 209,000 in 2000, 324,000 in 2005, 455,000 in 2009, 675,000 in 2019 and 935,000 in 2024. These older estimates, however, did not take into account downward adjustments of national estimates made by several countries such as the United States. The UN estimated in 2012, as a result of these adjustments, that there were only 316,600 centenarians worldwide. The following table gives estimated centenarian populations by country, including both the latest and the earliest known estimates, where available. A study which received a 2024 Ig Nobel Prize found these numbers are inflated by welfare and pension fraud and poor record-keeping, neither of which are uniform across jurisdictions.
== Prevention of nerve injuries == Methods to help prevent nerve injuries include injection pressure monitoring. The presence of a high opening injection pressure (> 20 PSI) is a sensitive sign of intrafascicular/intraneural needle tip placement. Extrafascicular needle tip placement is associated with low pressures (< 20 PSI). Also, high pressure injection was associated with neurologic deficits and severe axonal damage after the block. Other methods of preventing nerve injury include electrical nerve stimulation and ultrasonography. Electrical stimulation with a motor response at < 0.2 mA only can occur with an intraneural/intrafasciular needle tip location.

Cardarine is a common name for GW501516, a synthetic PPARδ agonist developed for research. It has not been approved as a medication in any country. It is classified as an unapproved drug and a prohibited substance in sport.
No. Cardarine is not an anabolic steroid; it belongs to a different chemical class that targets PPARδ. Steroids act primarily through androgen receptors, while cardarine acts through a nuclear receptor involved in metabolism.
Some rodent studies reported increased endurance and changes in fat metabolism after cardarine exposure. Long-term studies in animals also raised concerns about cancer in certain tissues. Human trial data are limited, so these findings cannot be directly translated to people.
It binds and activates PPARδ, a nuclear receptor that influences gene expression related to fatty acid metabolism and energy balance. This mechanism has been studied mainly in animals and cell models, not established as a safe human therapy.
